Abstract

hTNFalpha cDNA was inserted into retroviral vector to construct recombinant vector pL (TNFalpha) SN. The packaged and confirmed retrovirus were infected to RCC cell line. Then, nude mice were injected by 786-0 cell infected with the hTNFalpha gene or not, and the tumorigenecity of RCC was observed. The mean expressio level of TNFalpha in the supernatant of TNFalpha-transfected 786-0 cell-culture medium was (5 004±624)pg/ml. And the nude mice injected with TNFalpha gene engineered RCC cells had no tumor growth. The TNFalpha gene engineered human RCC cells lost tumorigenicity.
