Abstract
Objective: To observe the effect of Bushen Huoxue Tonglin Formula( BSHXTLF) on gene expression of Bax and Bcl-2 and ratio of Bax and Bcl-2 in Benign Prostatic Hyperplasia( BPH) rats. Methods: 72 rats were randomly divided into six groups: sham-operated group,model group,Proscar group,low dose group,middle dose group and high dose group.The rats model was established by injecting testosterone propionate 30 days after removing testis exclude sham-operated group. The drugs were administrated once a day at the same time. RT-PCR was used to measure the gene expression of Bax and Bcl-2,the calculating the ratio. Results: The expression of Bax mRNA in the treatment groups was increased compared with model group( P 0. 05). The expression of Bax mRNA in Proscar group,middle and high dose BSHXTLF group was increased compared with low dose BSHXTLF group( P 0. 05). The expression of Bax mRNA in high dose BSHXTLF group was increased compared with Proscar group and middle dose BSHXTLF group( P 0. 05). The expression of Bcl-2mRNA in the treatment groups was decreased compared with model group( P 0. 05). The expression of Bcl-2 mRNA in middle and high dose BSHXTLF group was decreased compared with low dose BSHXTLF group and Proscar group( P 0. 05). Ratio of Bax and Bcl-2 of each treatment groups was increased compared with model group( P 0. 05). Ratio of Bax and Bcl-2 of Proscar group,middle and high dose BSHXTLF group was increased compared with low dose BSHXTLF group( P 0. 05). Ratio of Bax and Bcl-2 of high dose BSHXTLF group was increased compared with middle dose BSHXTLF group and Proscar group( P 0. 05). Conclusion: BSHXTLF may increase gene expression of Bax and decrease gene expression of Bcl-2 and increase Ratio of Bax and Bcl-2. These are one of the mechanisms of BSHXTLF in the treatment of BPH.
