Abstract
Objective To explore the role and the possible mechanism of Diaomo Zhibengfang on the rats with endometria hyperplasia through observing the influence of the Diaomo Zhibengfang on apoptosis of endometrial cells and expression of bcl-2 and bax in the rats with ADUB.Methods Model of ADUB were induced by injection of oestradiol benzoate(0.15ml/100g) into castrated female healthy adult SD rats.Rats were randomly divided into four groups: control group,model group,big and small dose of Diaomo Zhibengfang.The model of ADUB was builded.After three days the rats in all groups were sacrificed.The expression levels of bcl-2 and bax were checked by TUNEL and immunohistochemistry.Results(1) Compared with control group,apoptotic index increased in the rats of the model group(P0.05).The apoptotic indexes of small and big dose of Diaomo Zhibengfang groups were higher than the model group(P0.05).There is no difference between small and big dose of Diaomo zhibengfang groups(P0.05).(2) The expression of bcl-2 increased,and the expression of bax decreased distinctly,bcl-2/bax ratio increased distinctly in the rats of model group(P0.05);(3) Compared with model group,the expression of bcl-2 was decreased distinctly of big and small dose groups,the expression of bax was increased,bcl-2/bax ratio decreased distinctly(P 0.05).Conclusion Diaomo Zhibengfang plays a role in the endometrial hyperplasia by accelerating the ecclasis of uterine endometrial.One of the mechanisms maybe related to inhibiting the expression of bcl-2 mRNA,enhancing the expression of bax mRNA,and decreasing bcl-2/bax ratio.
