Cluster buds induction and its regeneration system in Smilax riparia A. DC.

Abstract
The shoot tip of wild Smilax riparia A.DC was used as explants to conduct experiment on the induction,multiplication and rooting of cluster buds in media with different combinations of 6-BA(0.5-5.0 mg/L) and NAA(0.1-1.0 mg/L).The results showed that the optimum medium for cluster buds induction was MS+6-BA 2.0 mg/L+NAA 0.2 mg/L+ 0.3% activated charcoal.Browning of explants was completely controlled in this medium and the cluster buds grew earlier with 75.0 % induction rate.The optimum medium for multiplication of cluster buds was MS+6-BA 0.5 mg/L+NAA 0.1 mg/ L with 4.2 multiplication coefficient.The optimum medium for rooting was 1/2MS supplemented with 1.0 mg/L NAA,the rooting rate reached up to 87.0%,the number of root was from 4 to 7 and the average root length reached up to 1.2 cm.The rooting plantlets were found able to survive in the transplantation medium(nutrition soil ∶ sawdust ∶ yellow mud= 1∶1∶2) with 80.0% of transplant survival rate.
