Abstract
Objective To investigate the effect of endothelin-1 (ET-1) on hepatic ischemia-reperfusion injury in rats.Methods Eighty male Wistar rats were randomly divided into the normal control, ischemia-reperfusion, physiologic saline and endothelin-1 antibody groups and killed at 3 rd h after ischemia induced for 60?min and reperfused for 3?h. plasma and(or) liver (ET-1), glutamic-pyruvic transaminase enzyme (ALT), hyaluronic acid (HA), malondialdehyde (MDA) were measured and the histopathologically hepatic alterations were observed. Meanwhile, the changes of plasma ET-1 were also observed at the time points of 1, 3, 6, 12 and 24?h in the ischemia-reperfusion group. Results Plasma and liver ET-1, Plasma HA, plasma ALT and liver MDA concentrations were markedly increased after hepatic ischemia-reperfusion, whereas the plasma ET-1, HA, ALT levels in ET-1 antibody group were significantly decreased compared with the ischemia-reperfusion group(P<0.01, P<0.05). At the same time, the hepatic congestion and damages were markedly improved.Conclusion ET-1 might be participated in injury of the hepatic ischemia-reperfusion, which was correlated with the hepatic microcirculatory disturbances.
