Abstract
Objective To investigate the effect of endothelin 1 (ET 1) on hepatic ischemia reperfusion injury in rats.Methods Eighty male Wistar rats were randomly divided into the normal control, ischemia reperfusion, physiologic saline and endothelin 1 antibody groups and killed at 3 rd h after ischemia induced for 60?min and reperfused for 3?h. plasma and(or) liver (ET 1), glutamic pyruvic transaminase enzyme (ALT), hyaluronic acid (HA), malondialdehyde (MDA) were measured and the histopathologically hepatic alterations were observed. Meanwhile, the changes of plasma ET 1 were also observed at the time points of 1, 3, 6, 12 and 24?h in the ischemia reperfusion group. Results Plasma and liver ET 1, Plasma HA, plasma ALT and liver MDA concentrations were markedly increased after hepatic ischemia reperfusion, whereas the plasma ET 1, HA, ALT levels in ET 1 antibody group were significantly decreased compared with the ischemia reperfusion group( P 0.01 , P 0.05 ). At the same time, the hepatic congestion and damages were markedly improved.Conclusion ET 1 might be participated in injury of the hepatic ischemia reperfusion, which was correlated with the hepatic microcirculatory disturbances.
