Abstract

The tissue culture of Sagina saginoides from Tibet was studied systematically from aspects of disposal,shoot inducement,proliferation and rooting to transplantation.The results showed that in the proliferation period,6-BA was a major factor,the suitable medium was MS+6-BA0.5 mg/L+NAA 0.1 mg/L+30 g/L sugar,in which the propagation coefficient reached to 5.12,and the buds grew well,so the medium was suitable for transplantation.In the rooting period,the rooting rates of medium contained NAA and IBA were all lower than that of only contained IBA,the suitable medium was 1/2 MS+IBA 0.5 mg/L+20 mg/L sugar.In the transplantation period,the survival rate of plantlets with 2.5 mm of root length reached 100% in the mixture soil(leaf mold∶vermiculite as 3∶1).
