Determination of contents of total polysaccharide and gallic acid in the seeds of Castanea mollissima Blume by UV and HPLC

Abstract

Objective To determine the contents of total polysaccharide and gallic acid.Methods The contents of total polysaccharide were determined with ultraviolet spectrophotometry.The wavelength was set at 620 nm.Glucose was used as the control.The separation and quantitation of gallic acid was performed on a Diamonsil ODS C18 column(200 mm×4.6 mm,5 μm)using the mobile phase consisted of methanol-water-phosphoric acid(4.0∶96.0∶0.1)at 273 nm.The flow rate was 1 mL·min-1.Results The linear range of total polysaccharide and gallic acid was 45.0-225.0 μg(r=0.999 9)and 0.044-0.440 μg(r=0.999 8),respectively.The average recoveries of total polysaccharide and gallic acid were 97.7%(RSD 0.8%)(n=6)and 101.3%(RSD 1.2%)(n=6),respectively.Conclusions The methods can be used to control the quality of Castanea mollissima Blume.
