Abstract
Objective:Investigating the expressions of nerve growth factor(NGF)and interneukin-1 β(IL-1 β)after intracerebral hemorrhage(ICH)injury in rat brain and their regulation by Nao Yi An(NYA).Methods:VII type collagenase was stereotaxically injected into the right globas pallidus(GP)of rats at the coordinates of 1.4 mm anterior and 3.2 mm lateral to bregma and 5.6 mm ventral to the cortical surface to produce ICH animal model.Rats were executed by cervical dislocation at 1 d,2 d,4 d,and 7 d after ICH injury in batches.Northern blot and enzyme linked immunosorbent assay(ELISA)were employed to detect the mRNAs and proteins of NGF and IL-1 β in rat brain tissues.Results:The NGF mRNA was increased after 1 d of ICH injury,decreased to the level under normal expression at 4 d and 7 d after injury.But the NGF mRNA in NYA group still remained at the normal expression level after 4 d and 7 d of ICH injury.The IL-1 β mRNA level was sharply increased at 1 d and 2 d after ICH injury,still higher than normal expression at 4 d and 7 d after injury.In NYA group,the IL-1 β mRNA was markedly lower than in model group from 1 d to 7 d after injury.Both NGF protein IL-1 β protein contents were markedly increased in ICH model group and NYA treating group from 1 d to 7 d after in-jury.But the IL-1 β protein in NYA group was significantly lower than that in model group in all time points.Conclusion:The levels of NGF and IL-1 β mRNAs and proteins were increased after ICH injury in rat brain.NYA could markedly reduce IL-1 β expression and remain NGF expression.
