Abstract

Objective To observe the distribution of brain-derived neurotrophic factor (BDNF) mRNA in rats with intracerebral hemorrhage (ICH) and effect of Naoyi-an (NYA) on it.Methods The ICH rat models were established by microinjecting 0.4 l Ⅶ collagenase into the globus pallidus to observe the expression variation of BDNF mRNA at 2, 6, 12 h, 1, 4 and 7 d after ICH by in situ hybridization technique. And positive cell counts served as the observation index.Results BDNF mRNA expressed in cortex, hippocampus, striatum of normal controls and sham-operated rats. The expressions of BDNF mRNA in above-mentioned regions increased 2 h after ICH, but were not found in the hematoma region, and continuously increased in above-mentioned regions 6 h after ICH, then reached the peak after 1 d, began to decrease 4 d after ICH, and decreased continuously but were yet higher than the normal level after 7 d, and positive cells come around to the normal level in cells quantity and expression level 14 d after ICH . Both positive cells were mainly expressed in neurons. The BDNF mRNA expression level of NYA group were higher than that of the model group at 6 h, 1 and 4 d time point (P0.01). Conclusions NYA could upregulated the expression of BDNF mRNA after ICH.
