Abstract
Objective To investigate the effect of glucose at different concentrations on the cell cycle and p21 expression in cultured human peritoneal mesothelial cells(HPMC) and protection of puerarin.Methods HPMCs were cultured with serum free RPMI 1640 medium containing glucose of different concentrations(1.5% and 2.5%) and puerarin(100 mg/L) for 24 hours.The cell cycle distribution was then investigated by flow cytometry.The expression of p21 was detected by Western blot.Results In the presence of 1.5% and 2.5% glucose concentrations,most of the cells became hypertrophic,and were arrested in G1 phase of cell cycle.High glucose stimulated expression of p21,which had no difference between the 1.5% and 2.5%(P0.05).The number of the cells arrested in G1 and p21 expressions decreased in groups of puerarin.Conclusion High glucose stimulated the expression of p21,which might be related to the hypertrophy and arrest in the G1 phase of mesothelial cells by high glucose.Puerarin might antagonize the effect of high glucose of the cycles of mesothelial cells and their cyclin.
