Abstract
Objective:To investigate the effect of different concentrated glucose on telomere length in cultured human peritoneal mesothelial cell(HPMC) and Protection of Puerarin.Methods: HPMC were cultured with serum free RPMI 1640 medium containing different glucose concentrations(1.5% and 2.5%)and Puerarin(100μg.ml-1) for 24 hours.Iisolated the DNA,telomere length was detected by Southern blot.Results: In the presence of 1.5% and 2.5% glucose concentrations,most of the cells became hypertrophic.Telomere length of the groups of high glucose were shortened,and there were no difference between the 1.5% and 2.5%(P 0.05).Telomere length shortening were decreased in groups of Puerarin.Conclusion: High concentrated glucose stimulated the shortening of telomere length,which might be related to the hypertrophy.And Puerarin might resist the effect.
