Influence of high glucose on the expression of IL-18 and TGF-β1 in HBZY-1 rat glomerular mesangial cells

Abstract

Objective: To observe the expression of IL-18 and TGF-β1 in HBZY-1 rat glomerular mesangial cells and the relationship of them.Methods: Rat glomerular mesangial cells were cultured in normal glucose(NG group:the concentration of glucose was 5.5 mmol/L) and high glucose(HG group:the concentration of glucose 25 mmol/L) environments.Quanti-tative PCR were used to analyze the expression of IL-18 and TGF-β1 mRNA in each group.Then different concentrations of(IL-18 the concentrations of IL-18 were 1 ng/dL,10 ng/dL,50 ng/dL,100 ng/dL) were added and IL-18 antibody(the concentration was 100μg/dL) into each group.quanti-tative PCR were used to detect the expression of TGF-β1 mRNA in each group.Results: The levels of IL-18 and TGF-β1 in the HG group were higher than that in the NG group.The expression of TGF-β1 increased with the increase of function in the expression of IL-18.The expression of TGF-β1 was inhibited after blocking the expression of IL-18.Conclusion: Hyperglycemia can result in increased expression of IL-18 and TGF-β1.The increased expres-sion of TGF-β1 is dose-dependent on IL-18.Blocking the expression of IL-18 inhibites the expres-sion of TGF-β1.
