Abstract
Objective: To observe the change of negative cell-cycle regulatory protein p27 expression on glomerular mesangial cells under the influence of high glucose, ET-1 and IL-13. Methods: Glucose (5.5 mmol/L and 25 mmol/L), ET-1(10-7 mol/L) or IL-13(10 ng/ml)was added into the medium of cultured glomerular mesangial cells separately. The expression level of p27 was assayed by flow cytometry. Results: Compared with control group, the level of p27 in glomerular mesangial cells of glucose 5.5 mmol/L group at 48 h (5.14±0.94) and 72 h (3.84±0.81) decreased significantly (P0.001), glucose 25 mmol/L group (26.82±3.15 and 30.88±3.68) increased significantly (P0.05 at 72 h), of ET-1 group at 14 h and 18 h (14.76±1.49 and 12.18±1.30) decreased significantly (P0.05, P0.01), IL-13 group at 48 h (46.74±3.25) increased significantly (P0.001. Conclusion: Negative cell-cycle regulator p27 plays a very important role in regulating the proliferation of glomerular mesangial cells.
