Abstract
Objective: To investigate the role of pentadecapeptide BPC-157 in the proliferation,cell cycle,migra tion and tube formation of human umbilical vein endothelial cells(HUVEC).Methods: Different concentration of BPC-157(0,1,5,10,50,100 μg/mL) was adopted to stimulate HUVEC.The proliferation of HUVEC treated with BPC-157 was analyzed by MTT assay,and the percentages of HUVEC in G1,S and G2 phases were detected by flow cytometry.Wound scratch assay and transwell were designed to investigate the effect of BPC-157 on endo thelial cell migration.The effects on tube formation of HUVEC were evaluated by cavity forming experiment.Re sults: After the treatment with BPC-157 for 48 h,the rate of cell growth and the cell cycle of HUVEC have not changed.Compared with control group,BPC-157 could remarkably accelerate cell wound healing,and increase the number of migrated cells after 12 h.And the results clearly showed that BPC-157 treatment,especially 5 μg/mL dose,significantly promotes motility and tubular structure formation by HUVEC.Conclusion: Pentadecapeptide BPC-157 plays unimportant role in the proliferation and cell cycle of HUVEC,but it could obviously promote the migration and tube formation of HUVEC.
