Abstract

Objective To study the effect of THW-4,an extract from Trypterygium hypoglaucum on the proliferation of human umbilical vein endothelial cells (HUVEC).Methods HUVEC were cultured in vitro and different concentrations of THW-4 were added in experimental groups.Cell proliferation was evaluated by cell counting, MTT and and cell cycle detection.Results The inhibitory effects of THW-4 on proliferation of HUVEC displayed a dose dependent manner,with the halfinhibition concentration value (IC5)0 of 1.09 ng/mL at 24 h.Analysis of cell cycle indicated the THW-4 (0.5~10 ng/mL) mainly lead to the blockage of HUVEC in G0/G1 stage.Conclusion THW-4 has an inhibitory effect on the proliferation of HUVEC in vitro,and it has no protection on regeneration of functional endothelium after artery balloon.
