Study on Separation and Purification of Total Alkaloids and Mesaconine with AB-8 Macroporous Resin

Abstract

ATM: To systematically study the factors which affect separation and purification of the total alkaloids and mesaconine and optimize the purification technology.METHODS: The optimal conditions of absorption and elution were studied in the process of purifying two indexes with AB-8 macroporous resin by taking the content of the total alkaloids and mesaconine as indexes. RESULTS: AB-8 macroporous resin had the best separating efficiency when the mesaconine content in the extracted solution was 0.2 mg per 1 mL equivalent 1 g raw material, pH of the extracted solution was 10.0; the extracted solution was absorbed for 6 hours; then the resin was eluted using 7 times as much 95% ethanol as the volume of resin; AB-8 macroporous resin could used be five times repeatedly. The rate of extraction and purity of the total alkaloid was 80% and 30% or so respectively after the purification with AB-8 macroporous resin. CONCLUSION: AB-8 can meet industrial requirement.
