Abstract

Objective To explore the influence of Cs A on the maturation of cultured dendritic cell progenitors.Methods Bone marrow-derived DC progenitors were assigned to two groups:The cells in one group were cultured with grantdocyte-macrophage colony-stimulating factor(10 mg/L)and inter- lenkin-4(10 mg/L)for 13 days(control group),and those in other group with the same cytokines and CsA(1 mg/L)for 13 days(CsA group).Cell morphology was monitored under light and electron mi- croscopy and cell surface molecules including CDS0,CD86 and OX62 were assayed by flow cytometry.T cell proliferation was assayed by MTT.Results Compared with DCs in control group,DCs in CsA group were characterized as follows:(1)Lower expression levels of CDS0 and CD86(14.1%,17.1%);(2) Weaker activity of stimulating proliferation of allogenetic T cells.Conclusion The immature DCs inhib- ited by CsA may have the properties of inducing allogenetic-specific immune inhibition in vitro.
