Abstract

Objective: To explore the influence of CsA on the maturation of cultured dendritic cell(DC) progenitors. Methods:Bone marrowderived DC progenitors were assigned to two groups: in control group,DC was cultured with granulocyte-macrophage colony-stimulating factor(10 mg/L) and interleukin-4(10 mg/L) for 13 days(control group),in CsA group,DC was cultured with the same cytokines and CsA(1mg/L) for 13 days(CsA group).Cell morphology was monitored under light microscopy and cell surface molecules including CD80,CD86 and OX-62 were assayed by flow cytometry.T cell proliferation was assayed by MTT. Results:Compared with DC in the control group,DC in CsA group were characterized as follows:①lower expression levels of CD80 and CD86(14.1%,17.1%);②Weaker activity of stimulating proliferation of allogenetic T cells. Conclusion:The maturation of DC inhibited by CsA may have the properties of inducing allogenetic-specific immune tolerance in vitro.
