Effects of Cyclosporine A on Murine Bone Marrow-Derived Dendritic Cells

Abstract

OBJECTIVE To study the effect of cyclosporine A(CsA) on murine bone marrow-derived dendritic cells in vitro.METHODS Murine bone marrow-derived dendritic cells were co-cultured with CsA at the doses of 0,0.5 and 1 mg·L~(-1).Immunophenotype of DC was analyzed by flow cytometry.The stimulatory activity of DC on allogeneic T cells were evaluated by the incorporation of()~(3)H-TdR in mixed lymphocyte reaction.Cytokines such as IL-12P70,TNF-α and IL-10 in culture supernatant were detected by ELISA assay.RESULTS CsA inhibited the expression of surface costimulatory molecules CD80 and CD86,increased the expression of BT-DC,B7 family new member,but showed no effect on B7-H1.CsA-treated DC exhibited significant impaired ability to stimulate the proliferation of allogeneic T cells with a reduction of IL-12P70 and TNF-α production and a higher level of cytokine IL-10.CONCLUSION Murine bone marrow-derived dendritic cells have the properties of immature dendritic cells and induce immunologic tolerance under the direct action of CsA via the down-regulated expression of CD80 and CD86 costimuLatory signal and the up-regulated(expression) of B7-DC inhibitory signal.
