Compound formula Jiukele′s efficacy and its mechanisms in prevention and treatment of experimental alcoholic liver fibrosis in rats

Abstract

Objective:To investigate the efficacy of Chinese compound formula Jiukele in the prevention and treatment of ethanol-induced liver fibrosis in the rats and its mechanisms.Methods:Forty male Wistar rats were divided randomly into 5 groups: normal control group,model group,therapy group,high and low dose prophylactic group.The mixture of alcohol(55.),pyrazole and olive oils was fed to the stomach of the rats of model group every morning(7∶ 00-8∶ 00).The rat model was successfully established in 12 weeks.The Jiukele was given to the rats of therapy group for another 4 weeks after rat model was established successfully.In high and low dose prophylactic group,the mixture was fed to the rats every morning and Jiukele was fed to the rats after 2 hours.The experiment was over in twelve weeks,except the therapy group,blood samples and liver tissues were collected.The samples of therapy group were collected after additional 4 weeks.The weight of the rats,liver function,the content of serum PⅢP,LN,the morphology and the lever of TGF-β1 in liver tissure were observed.Results:In model group,the content of serum ALT,AST,PⅢP,LN,the weight of rats,the pathology morphology of liver tissures,the expression lever of TGF-β1 in the liver tissure were significantly higher than normal control group(P0.01).The weight of the rats in high dose prophylactic group was almostly increased the same as the normal control group(P0.05).In high and low dose prophylactic group and the therapy group,the content of the serum ALT,AST,PⅢP,LN were obviously lower than the model group(P0.01).The contents of TGF-β1 in the liver tissures of model group was greatly higher than other groups(P0.01),the pathology morphology change was also more serious in liver tissues of model group than other groups.Conclusion:Jiukele has effects in prophylaxis and the treatment of alcoholic liver fibrosis in rats.The mechanisms may be as follows:to suppress the deposition and accelerate degradation of liver extracellular matrix,alleviate the pathology morphology change of liver tissure;to improve liver function,immune function,adjust liver metabolism disorder.
