Expression of ICAM-1,TNF-α,IL-6,LPS on Severe Acute Pancreatitis Associated Gut Barrier Dysfunction in Rats

Abstract
Objective To investigate the process of severe acute pancreatitis associated gut barrier dysfunction,and to explore the early diagnosis basis of gut barrier dysfunction.Methods Rats were randomly divided into groups SAP and groups control.Enzyme linked innunosorbent assay(ELISA) were separately used to detect the serum levels of TNF-α,IL-6,LPS of both group,the expression of ICAM-1 of both group were measured by strept avidin-biotin comple-αlkaline photosphatase separately,and the bacterial culture of samples from pancreas,liver,and mesentery were made separately,and changes in pathology were observed.Results Compared with control groups,the serum levels of TNF-α,IL-6,LPS,AMY and the expression of ICAM-1 in the intestinal mucosa increased significantly in SAP groups(P0.05),also significantly different when compared with pancreas pathology,there were positive correlations with the expression of ICAM-1 when compared with the serum level of LPS(P0.01) and pancreas pathology(P0.01) as time goes by.The bacterial culture of pancreas,liver and mesentery all were positive in SAP groups and also there was positive when compared with control groups.Conclusions The high expression of ICAM-1 and the serum levels of LPS show it tightly corresponded with severe acute pancreatitis associated gut barrier dysfunction;the serum levels of TNF-α,IL-6,LPS with severe acute pancreatitis associated gut barrier dysfunction are important during the disease;and the intestinal barrier dysfunction probably lead to enteric bacterial translocation and endotoxemia.
