Effects of high glucose on β1-integrin expression and extracellular matrix secretion in glomerular mesangial cells

Abstract

Objective To observe the effects of high glucose on β1 integrin mRNA expression in cultured glomerular mesangial cells ( GMC). Methods Cultured glomerular mesangial cells were treated with glucose in different concentrations. The proliferation of GMC was measured by MTT, the expression of β1 integrin was estimated by RT-PCR, and the secretion of fibronectin and laminin were measured by ELISA, respectively. Results High glucose inhibited the proliferation of GMC and promoted the expression of β1 integrin, which had a significant correlation with the secretion of fibronectin and laminin. The effect was dose-dependent. Conclusion Hyperglycemia, as an initial factor of diabetic nephropathy, may increase the synthesis and secretion of fibronectin and laminin of the extracellular matrix (ECM) through the up-regulation of β1 integrin mRNA expression.
