Effects of High Glucose Solution in Secretion of Extracellular Matrix of Rat Glomerular Mesangial Cells in Vitro

Abstract

Objective:To observe the effects of high glucose solution on secretion of extracellular matrix(ECM)of cultured rat glo-merular mesangial cells. The levels of fibronectin(FN), laminin( LAM), type Ⅳand type Ⅲ collagen levels in the tissue cul-tured supernatant were measured. Method: The culture of rat glomerular mesangial cells was established and the FN, LAM, typeⅣ and type Ⅲcollagen were analysed by a solid-phase enzyme-linked immunosorbent assay (ELISA). Results: High glucose solution selectively affected a group of function-related proteins. Conclusion: In vitro high glucose solution increased mesangial cell synthesis of extracellular matrix proteins. The present studies suggest that mesangial cells may increase ECM synthesis in response to hyperglycemia and the response could contribute to the accumulation of extracellular matrix to pro-duce diabetic glomerulosclerosis.
