Abstract
Objective To observe effects of high gluose condition on expressions of hyaluronic acid(HA),laminin(LN)and Ⅳ type collagen in extracellular matrix(ECM) secreted by glomerular mesangial cells(GMC),therefore provide experimental evidence for diagnosis and clinical treatment on diabetic nephropathy.Methods (1)The levels of HA,LN and Ⅳ type collagen in the tissus culture supernatants were examined by a solid phase enzyme linked immunoadsorbent assay(ELISA);(2)Effects of different concentrations of glucose on glomerular mesangial cell proliferation were detected by MTT method. Results High concentration of glucose inhibited the proliferation of mesangial cells,which was positive correlative with glucose concentrations.The levels of HA,LN and Ⅳtype collagen increased largely between 48 to 72 hours after culture on high concentration of glucose.The high levels lasted for a week.Increasement of ECM was independent on cell proliferation.The control group was in a stable condition.These results showed high glucose may play an important role in promoting ECM′s excretion by GMC.Conclusion High concentration of glucose may restrain GMC proliferation in a concentration dependent manner.Glomerular mesangial cells may increase ECM synthesis on high concentration of glucose.The accumulation of extracellular matrix may contribute to the pathogenesis of diabetic nephropathy.
