Abstract
Objective:To observe the effects of Wenzhong Jianpi Decoction on 5-HT expressions in hypothalamus and colon mucosa of rats of diarrhea-predominant irritable bowel syndrome(D-IBS).Methods: Forty-eight Wistar rats were randomized into four groups: normal group,model group,herb group and control group,12 rats in each group.D-IBS model was made by intragastric administration of senna leaf and binding stress.After 2-week intervention,the 5-HT expressions in hypothalamus and the colon mucosa were assayed by IHC method and then analyzed by a special images analysis system.Results: The positive area(PA),optical density(OD) and IHC-I in the hypothalamus and the colon mucosa were significantly higher in model,control and herb groups than in the control group(P0.05,P0.01);and they were strikingly lower in herb and control groups than in model group(P0.05,P0.01);except for PA in the hypothalamus,other indicators were lower in control group than in herb group(P0.05,P0.01).Conclusion: Wenzhong Jianpi Decoction can reduce 5-HT expressions in the hypothalamus and colon mucosa in D-IBS rats to regulate brain-gut axis,and hence to treat D-IBS.
