Expression of Inflammatory Cytokine in Colon Mucosa of D-IBS Rats and Effects of “Wenzhong Jianpi Decoction” on Them

Abstract

Objective:The aim of our study was to detect the expression of IL-1β,IL-10 in the colon mucosa of rats and then investigate the effects of Chinese herbs on them. Methods:Totally 60 rats were divided into four groups,and the traditional method for irritable bowel syndrome and intragastric administration of Folium Cassiae were combined to develop the IBS models,then we determined the contents of IL-1β,IL-10 by IHC methods(Envision)and analyzed the positive area(PA),optical density(OD)and IHC index(IHC-I). Results:Our study showed that in the model rats,and also in the rats treated by Chinese herbs and western medicines,the IL-1β expression including PA,OD and IHC-I showed a decreasing trend than the normal rats,and there were significant differences between the model group,western medicine group and the normal rats. While IL-10 was decreased significantly in the model rats than that in the normal group,including PA,OD and IHC-I,the same results were seen in the both treatment groups. After treated with Chinese herbs,the PA,OD and IHC-I were raised significantly;this can be seen in the group which was treated with western medicine too,and approximately with the normal rats,although PA did not change in this group. Conclusion:It is thought that there was a disequilibrium between inflammatory factors and anti-inflammatory factors during the development of IBS exactly,and the decrease of IL-10 plays an important role in this course. Due to the decline of IL-10,there was low grade inflammation in the colon mucosa persistently. Chinese herbs can promote the expression of mucosal IL-10 and media the equation of IL-1β/IL-10,enhance the anti-inflammatory ability of colon mucosa,thus the curative effects can be received.
