Effects of Changjitai Decoction on the Changes of Colon Mast Cells and the Expression of 5-hydroxytryptamine in Rats with Irritable Bowel Syndrome of Diarrhea Type

Abstract
Objective To observe the changes of mast cells(MCs) and the expression of 5-hydroxytryptamine(5-HT) in colonic mucosa of rats with irritable bowel syndrome of diarrhea type(IBS-D),and to explore the mechanism of Changjitai Decoction,a traditional Chinese medicine,in treating IBS-D.Methods Sixty male Sprague-Dawley rats were averagely randomized into 6 groups: normal group,model group,western medicine(dicetel) treated group,and large-,middle-and small-dose ChangjitaiDecoction treated groups.Except for rats in the normal group,the other rats were induced to IBS-D model by using restraint stress,Radix et Rhizoma inducing diarrhea,and colorectal distension,then the IBS-D rats were treated with corresponding drugs,respectively.The frequencies and characteristics of feces were observed in all rats during the experiment.After the rats were sacrificed,their colon specimens were collected for observing the counts,the area,and the ratio of nucleus to plasma of MCs and for determining the contents of 5-HT in the colon mucosa.Results Compared with those in model group,the density and the area of MCs were decreased,the ratios of nucleus to plasma of MCs were increased,and the contents of 5-HT were down-regulated in all treatment groups.Among them,the high-and middle-dose Changjitai Decoction treated groups and dicetel treated group showed a significant difference(P0.05,or P0.01).The characteristics of feces were significantly improved in high-dose Changjitai Decoction treated group and dicetel treated group(P0.05).But there was no significant difference in the improvement of feces characteristics between Changjitai Decoction treated groups and dicetel treated group.Conclusion Changjitai Decoction can ameliorate visceral hypersensitivity,and regulate restrain stress induced disorders of intestinal dynamics by regulating the stability of MCs and the contents of 5-HT in rat colonic mucosa.
