Observation of short-term efficacy of nedaplatin combined with 5-fluorouracil and leucovorin in treatment of advanced esophageal carcinoma

Abstract

Objective To compare short-term efficacy and toxicity between nedaplatin(observation group)and cisplatin(DDP)(control group) in combination with 5-fluorouracil(5-Fu)and leucovorin(CF) in treatment of advanced esophageal carcinoma.Methods Sixty-eight advanced esophageal carcinoma patients who had been proved by pathology or cytology were treated with combined chemotherapy.There were 36 patients in observation group and 32 patients in control group.The 2 groups were comparable.Nedaplatin,100 mg/m2,iv drip,on day 1;5-Fu,375 mg/m2,iv drip,from day 1 to 5;CF 200mg/m2,iv drip from day 1 to 5;DDP 25 mg/m2,iv drip from day 1 to 3.21 days contituted a cycle.All patients were treated for 3 cycles.Results The total effective rate in observation group was 55.6%,while control group was 46.9%,without significant difference between them(P0.05).The incidences of gastroenteric and kidney toxicity were markedly higher in control group than in observation group(P0.05).Conclusions The short-term therapeutic efficacy between the DDP regimen and nedaplatin regimen was similar,but the incidence of gastroenteric and kidney toxicity of DDP regimen was significantly higher than nedaplatin regimen.
