Abstract

[Objective] To choose the most suitable induced melon Huangzuixian leaves callus and adventitious bud medium,and suitable for the adventitious bud elongation and rooting medium.[Method] With melon Huangzuixian seed as the materials,cotyledon as explants,adding different concentration of BA and IAA hormone combination of the MS medium on the callus and adventitious bud induction,adventitious bud elongation and the effects of adding different concentration IBA hormone combination of the MS culture medium on the rooting effect were studied.[Result] The most suitable induced melon Huangzuixian leaves callus culture medium is MS + 0.5 mg/L BA + 2.0 mg/L IAA and MS + 1.0 mg/L BA + 2.0 mg/L IAA,the most suitable for its cotyledon adventitious bud induction medium is MS + 0.5 mg/L BA + 0.5 mg/L IAA,adventitious bud elongation of the most suitable medium is MS + 1.0 mg/L IAA rooted appropriate medium is 1/2 MS + 1.0 mg/L IBA.[Conclusion] A culture medium which is suitable for melon Huangzuixian leaves regeneration system was selected.
