Abstract

【Objective】 The objective of this study was to optimize the extraction conditions with cellulase pretreatment of isoflavones from pueraria root residue.【Method】 Single-factor test and response surface methodology(RSM) were used to analysis the effects of the enzyme dosage,enzymolysis temperature,pH value and enzymolysis time on the extraction yield of isoflavones.【Results】 The optimal enzymolysis conditions were got through the regression model,which are cellulase dosage 11 mg per 5 g pueraria root residue,enzymolysis temperature 51 ℃,pH 5.0,enzymolysis time 2.3 h.Under these conditions,the extraction yield of isoflavones can reach at 12.34 mg/g,which is increased 57% than that of traditional method.【Conclusion】 The result shows that the optimal conditions with cellulase pretreatment can effectively improve the extraction amount of isoflavones from pueraria root residue.
