Abstract

The factors influencing plant regeneration of the callus from the mature embryo of Ryegrass which including explant sterilization, callus induction, subculture and plant regeneration was studied. The result showed that the explants, surface-sterilized by a solution of 75% NaClO(7%-10%active chlorine)for 25-30 min after dipped in 70% ethanlo for 1 min could get optimam sterilization effect. The feasible medium for induction of callus was MS. basal medium supplemented with 2.0-(3.0 mg/L) 2,4-D, 0.1 mg/L 6-BA, in which the rate of the callus induction reached to 86.67%.
