High Frequency Plant Regeneration from Mature Embryo of Wheat

Abstract

Aiming to setting up an effective wheat high frequency regeneration system,the mature embryo of wheat variety Ganchun 24 was used as exophyte to explore the changes of callus induced and the influence of differentiation during the treatments of different disinfection way,including different soaking seeds time,different means of inoculation,and different 2,4-D concentration.The results showed that different disinfection way,2,4-D concentration and different means of inoculation had significant effects on differentiation of wheat callus of mature embryos;but different soaking seeds time had no significant effects.Using the 25% NaClO 25 min on the first disinfection,after soaking seeds utilizing 25% NaClO 10 min,the method could have good effects on callus induced and had less damage on the condition of lower rate of infection;soaking seeds of 15-21 h had no significant effects on the callus induced of wheat mature embryos,but 18 h or 21 h was better to the quality of callus induced;scraping the mature embryos was helpful for callus induction and differentiation,the rate of induction and differentiation were 98.33% and 62.04% respectively.When the 2,4-D concentration was 2.5 mg/L,the quality of embryonic callus was better of Ganchun 24.
