Abstract
Objective:To establish a standard process of measuring anti-cancer drugs sensitivity with MTT assay by optimizing the experiment.Methods:Some influencing factors of MTT assay were used in studying the sensitivity of K562 cell lines to anti-cancer drugs carboplatin and paclitaxel.Results:A better result was obtained in the condition of each well contained 5×103 K562 cells,anti-cancer drugs continuously contacted with the cells for 48h,20μl MTT solution(5mg/ml) was added and incubated for 6h,and the optical density at 570nm single wavelength was measured.Conclusion:A standard MTT assay for anti-cancer drugs sensitivity test in vitro might provide experimental basis for clinical individual chemotherapy.
