Influencing Factors of the MTT Based on Tumor Chemosensitivity Assay in Vitro(A Study Report)

Abstract

Objective:To establish a standard process of measuring the anti-cancer drug sensitivity with MTT assay by analyzing the influencing factors of the chemosensitivity assay in vitro.Methods:Some influencing factors of MTT assay were found in studying the sensitivity of human peripheral blood mononuclear cells(PBMC) to anti-cancer drugs Oxaliplatin and Paclitaxel and the time of selective culture and MTT incubation time were optimized.Results:It showed that a better result was obtained in the condition with each containing 4.0×104 PBMC,the anti tumor drugs continuously contacted with the cells for 48 h,20 μL MTTwas added and incubated for 6 h,the optical density at 570 nm single wavelength was measured.Conclusion:A kind of standard MTT assay for drug sensitivity test in vitro might provide experimental basis for clinical individual chemotherapy.
