Experimental Study on the Anti-cancer Drugs' Sensitivity in the Colormetric Assay of Cancer Cells with MTT and Its Effective Factors

Objective To study the anti-cancer drugs' sensitivity in the colormetric assay of cancer cells with MTT and its effective factor.Methods 45 primary cancer cell patients in vitro (15 samples of gastric cancer, 10 samples of lung cancer, 12 samlples of colon cancer, 8 samples of breast cancer) were examined with MTT assay to find out their drugs' sensitivities to the often-used chemotherapy medicine.Results The sensitivity of the 4 kinds of cancer cells to the 8 kinds of chemotherapy medicine is different.Using 30 minute digestion time, 100%～60% concentration of lymphocyte seperating liquid and doing the assay as soon as the sample obtained,the percentige of 94 65±28 47, of the living cell,is significant different with the other experimental condition.Conclusions MTT assay is an easy and quick method for drug-sensitivity test in vitro and provides experimental proof for individual chemotherapy.Digestion time,concentration of lymphocyte seperating flud and the time of sample storager may be important factrs for the results of the assay.
