Abstract
A high performance liquid chromatographic method with microcolumn for the determination of rutin and quercetin in buckwheat(fagopyrum tataricum) was studied.Rutin and quercetin were extracted from the sample by refluxing with 90% methanol for 30 min.Rutin and quercetin in the extract were separated on Waters Xterra~(TM) RP_(18)(1.0 mm×50 mm,2.5 μm) chromatographic column with methanol and 1% acetic acid(60∶40) as the mobile phase passing through the column at a flow-rate of 0.2 mL·min~(-1),and detected with the photodiode array detector at 360 nm.In the analysis of 4 buckwheat samples by this method,RSD′s obtained were in the range of 0.51%~1.20%,and recoveries in the range of 98%~102%.
