Abstract
[Objective]The aim was to research the method of high-quality DNA extraction from leaves tissue culture of the tetraploid Isatis.[Method]The extraction effects of five different DNA-extracting methods such as CTAB method 1,CTAB method 2,SDS Act 1,SDS France 2 and the alkaline lysis method were compared by the UV spectrophotometry and agarose gelelectrophoresis.[Result]The results showed that CTAB method 2 was the best method,OD260/OD230 value was 2.311,OD260/OD280 value was 1.774,DNA was the most pure,and it had the largest concentration was the highest rate of extraction and the best DNA gua lity,PCR bands amplified clear and reproducible.[Conclusion]CTAB method 2 was an effective way for DNA extraction from leaves tissue culture of the teproducible Isatis.
