Comparison of Genomic DNA Extraction Methods From Hyacinth

Abstract

Five methods including simple CTAB,CTAB improved method,SDS-CTAB method,high salt precipitation method and silica-purification method were used to extract genomic DNA from different parts of Hyacinth such as bud,leaves,scale.The results were compared by three parts,purity,the electrophoreses results and yield of the extracted DNA.The results showed that all of these DNA extraction methods except for CTAB-silica-purification method could obtain DNA.The purity of extracted DNA was as following : simple CTAB method SDS-CTAB method CTAB improved methodhigh salt precipitation method,and yield as following : simple CTAB methodCTAB improved methodhigh salt precipitation methodSDS-CTAB method.Simple CTAB method could obtain more purity DNA than the other methods and OD260/OD280 was 2.01,OD260/OD230 was 2.33.The concentration was 406.8ng·L-1 and the yield was 175.95ng·g-1.The extraction results from different parts showed that flower buds and leaves were both suit for Hyacinth obtain DNA.
