Abstract
Using the seed endosperm of Pinus armandi Franch as experiment material,the genomic DNA of Pinus armandi Franch was extracted using three methods,which were the classical CTAB method,simple CTAB method and SDS micro-extraction method.By agarose gel electrophoresis,ultraviolet spectrophotometer,standard deviation and standard error of arithmetic mean,Chi-square test and SPSS,the advantages and disadvantages of three methods in the yield,quality and stability of extracting DNA were summerized.The results showed that by the Chi-square test,the differences of the data caused by the method,not the accidental error.Compared with the classical CTAB method,by the(OD260-OD320)/(OD280-OD320),the standard deviation and standard error of SDS micro-extraction method and simple CTAB method were little different.The data and results were relatively precise.By the SPSS,the simple CTAB method with other two methods had significant differences.The SDS micro-extraction method and classical CTAB method had not significant differences(P 0.05).The extraction effect difference was not significant.In conclusion,compared with the simple CTAB method,classical CTAB method and SDS micro-extraction method had the rate of(OD260-OD320)/(OD280-OD320) closing to 2.0 and might have RNA pollution with low DNA concentration,so classical CTAB method and SDS micro-extraction method were not suitable for extracting endosperm genomic DNA in the P.armandi Franch.The simple CTAB method had the rate(OD260-OD320)/(OD280-OD320) closing to 1.8 and had no protein and RNA pollution with the highest concentration and purity of DNA,so the simple CTAB method was suitable for extracting endosperm genomic DNA in the P.armandi Franch
