Abstract
Objective To investigate the mechanishs of Kanglaite enhancing the inhibitory activity of Adrimay (ADR) on HL-60 cells. Methods ① HL-60 cells at logarithmic growth phase were treated with different concentration of Kanglaite for various time period, The growth curve of HL-60 cells was observed by cell count. Changes of cell cycle were analyzed by flow cytometry. ②After kanglaite treated HL-60 cells for 96h, The inhibitory effects of ADR with certain concentration on HL-60 cells were analyzed by MTT assay, The intracellular ADR concentration was detected by flow cytometry. Results ① The HL-60 cells treated by 1mg/ml Kanglaite for 6 days, The cell count are (10.3±1.1)×10 5个/ ml,compare with control group's (20.5±3.5)×10 5个/ml, the inhibitory effects are achieved with statistical significance, P0.05. ② The HL-60 cells treated by 1mg / ml Kanglaite for 96h, The analysis of cell cycle shows kanglaite can increase the amount of cells in S phase (64.76±1.2)% ,compare with control group's (56.67±0.6)% P0.05. ③ After 0.1mg/ml kanglaite treated HL-60 cells for 96h, The inhibitory effect of 0.001μg/ml ADR-treated for 24h (24.2±5.3)% are statistical higher than that of control group(16.7±3.9)%, P0.05. The intracellular ADR fluorescence of 0.2μg / ml ADR-treated for 6h(12.8±1.7) are statistical differences with control group's(8.8±1.2),P0.05. Conclusions HL-60 cells exposed to Kanglaite resulted in dose-depentent inhibitory, It's mechanishs may be kanglaite retarding HL-60 cells passing S phase, and enhancing the intracellular ADR concentration of HL-60 cells.
