Effects of Yishen Tongluo Decotion on PAI-1 Expression of Membranous Nephropathy Rats

Objective:To observe the effects of Yishen Tongluo Decotion on the expression of PAI-1 of membranous nephropathy rats.Methods:SD rats were randomly divided into 4 groups:control group,model group,western medicine group and Chinese herb group(n=10,in each group),each rat was given cationic bovine serum albumin(C-BSA)1mg,adding 0.5mL phosphate buffer Night(PBS),and the same amount of Freund's incomplete adjuvant was mixed into a milky white suspension,double sides of the armpit and multi-points of groin were injected pre-immunized.1 week later,each rat tail vein was injected of C-BSA2.5mg,3 times a week for 3 weeks.After the modeling,the normal group and model group were fed once daily administration of a considerable measure of saline for 4 weeks.Model group:fed once a day administration of a considerable dose of saline;benazepril group:benazepril tablets ground into powder,dissolved in 5mL normal saline at 10mg/kg daily gavage once,for 4 weeks.Kidney Tongluo group:fed fried Yishen Tongluo decoction 5mL,once a day for 4 weeks.At the seventh week,24h urine protein quant(UPro),TP,ALB,the electron microscope observation、the immunofluorescence observation and PAI-1 were detected with relative techniques.Results:Compared with normal group,the 24h urinary protein quantitative analysis and the levels of TC and TG of the blood serum of model group were sharply increased.TP and Alb of the blood serm were significantly lower than normal group,nephritic injury was incured.The expression of PAI-1 in model group was increased(P0.05).Compared with model group,the 24h urinary protein quantitative and the levels of TC and TG of the blood serm of each treatment group were sharply decreased,TP and Alb of the blood serm were significantly higher than model group.Nephritic injury was lighter.The expression of PAI-1 in model group was increased as compared to the Benazepril and Chinese herb group(P0.05).There was no markly difference between the Benazepril and Chinese herb group(P0.05).Conclusion:Yishen Tongluo Decotion can reduce rat's proteinuria,increase plasma protein,improve blood rheology to protect renal function.Yishen Tongluo Decotion can depress PAI-1 expression in nephridial tissue.
