Optimization of the extraction of Cordyceps Militaris polysaccharides by microwave assisted with enzyme hydrolysis

Abstract

To optimize the microwave-enzyme synergistic extraction process of Cordyceps militaris polysaccharides from abandoned Cordyceps militaris medium,on the base of single factor investigation,effects of cellulase content,enzymolysis time,microwave power,and microwave extract time as well as their interactions on yield of the polysaccharides were studied by response surface methodology( RSM) analysis.The optimal extraction conditions were obtained as follows: ratio of solid- liquid 1 ∶ 30,the amount of cellulase 1650U /g,enzymolysis temperature55℃,pH5.5,enzymolysis time 44min,microwave power 480W,extraction time 3.5min.Under the optimal condition,the yield of Cordyceps militaris polysaccharides was up to 18.45%.
