Abstract
Objective To investigate the effect of Gadolinium chloride(GdCl3)on acute liver injury by Galn/LPS.Methods GdCl3(10 mg/kg)or the same volume of 0.9% Sodium Chloride was continually injected into tail vein at 48 h and 24 h before Galn/LPS intraperitoneal injection. The blood was collected from angular vein and liver was extirpated with barbital anaesthesia after LPS or 0.9% Sodium Chloride injected at 0.5,1,1.5 h and 6 h for assaying ALT,GSH,MDA and TNF-α.Results Galn/LPS-induced acute liver injury was attenuated significantly by GdCl3 pretreatment. The plasma ALT activity and the content of MDA and TNF-α in liver decreased remarkably by GdCl3 pretreatment while the content of GSH in liver increased significantly. Conclusion GdCl3 pretreatment significantly inactivated KCs and decreased TNF-α and ROS induced by LPS,therefore attenuated liver injury.
