Effect of Tonifying Spleen and Nourishing Liver Decoction on VEGF Expressions in Tumor Tissues of Mice Bearing Hepatoma H22

Abstract

Objective:To investigate the anti- tumor effect and its mechanism of Tonifying Spleen and Nourishing Liver Decoction in hepatoma H22 mice. Methods:Fifty BALB /c mice bearing hepatoma H22 were divided into 5 groups:saline(NS)group,the low,moderate,high dose with Tonifying Spleen and Nourishing Liver Decoction and 5-Fluorouracil(5-FU) groups. Inhibition of Tonifying Spleen and Nourishing Liver Decoction on tumor was investigated. The impact of Tonifying Spleen and Nourishing Liver Decoction on expression of vescular endothelial growth factor mRNA in tumor tissues of hepatoma H22 bearing mice was investigated by quantitative real- time polymerase chain reaction(RT- PCR). Results:The inhibitory rate of tumor in hepatoma H22 bearing mice in low,moderate and high dose groups and 5- FU groups was20. 56%,50. 58%,23. 34% and 77. 52%,respectively. The weight of tumor in moderate dose group and 5- FU group was significantly different with that of saline control group( P 0. 05) and the weight of tumor in low and high dose groups had no difference with that of saline control group(P 0. 05). The middle and high dose of Tonifying Spleen and Nourishing Liver Decoction could significantly down- regulate the expression levels of VEGF mRNA in tumor tissues from the mice bearing Hepatoma H22(P 0. 05). The effects of the mid dose groups were the most prominent and showed significantly difference in statistics(P 0. 01). Conclusion:Results indicate that Tonifying Spleen and Nourishing Liver Decoction has notable anti- tumor effects and the possible mechanism of it inhibiting proliferation was associated with down- regulating the expression level of VEGF mRNA in tumor tissues.
