Chinese medicine with replenishing qi and invigorating spleen effects on protein kinase C activity in liver and kidney of nude mice with gastric cancer

Abstract

Objective To discuss the effect of Chinese medicine on protein kinase C(PKC)activity in liver and kidney of nude mice with stomach cancer.Methods Thirty nude mice out of 40 were selected randomly and armpit inoculated with human gastric cancer cells SGC-7901. The nude mice were randomly divided into model group,tonifying spleen control group and tonifying spleen anticancer group,with 10 mice in each group. The rest of 10 mice were divided into blank group. The mice in blank group and model group were treated with normal saline,tonifying spleen control group was treated with Chinese medicine with replenishing qi effect,tonifying spleen anticancer group was treated with Chinese medicine with combined replenishing qi and anticancer effects,with 0. 8 ml each and for 14 days by gavage. The condition of the liver and kidney tissue were observed and the weights were measured. The organ coefficients were counted and the PKC activities in liver and kidney were tested by Western Blot method. Results Compared with blank group,the liver coefficient and kidney coefficient in model group were decreased,with significant difference between the two groups(P 0. 05); compared with model group,the liver coefficient and kidney coefficient in tonifying spleen control group and tonifying spleen anticancer group were increased significantly(P 0. 05). The PKC activities in liver and kidney of the model group were significantly higher than that of the blank group. After treatment,the PKC activities in liver and kidney of the tonifying spleen group and tonifying spleen anticancer group were lower than that of the model group,and the PKC activity in tonifying spleen anticancer group was lower than that of the tonifying spleen control group significantly(P 0. 01). Conclusion Spleen deficiency symptom is very common in mice with gastric cancer. Chinese medicine has regulatory effect on the body weight,liver weight,kidney weight,liver coefficient,kidney coefficient and PKC activities in nude mice. Moreover,the Chinese medicine with replenishing spleen qi and anticancer effects has better regulatory effect on PKC activities than that with replenishing spleen qi effect.
