Abstract

Objective To study the effect of formaldehyde inducing DNA-protein crosslinks(DPC) and DNA damages in mice lung.(Methods)Mice lung was taken as experimental material,KCl-SDS assay and comet assay were used to determine the amount of DNA-protein crosslinks and effect of DNA damages induced by liquid formaldehyde in vitro.(Results) KCl-SDS assay showed that formaldehyde could not cause DPC at lower level(5 μM,25 μM,P0.05),while at higher level(125 μM,P0.05;625 μM,P0.01) it caused DPC obviously.According to comet assay,it caused DNA strand breakage(DSB) at low level(5 μM,25μM,P0.01) and caused crosslinks at higher level 125 μM,625 μM,P0.01).Conclusion It was considered that formaldehyde might cause DPC at higher level,but at lower level(25 μM),it mostly caused DSB.
