Abstract
Objective:To establish methods for identification of adenosine and adenine in Ganoderma capsules by TLC and determination of adenosine by HPLC.Methods :TLC,the stational phase was GF_(254),the mobile phase con- sisted of chloroform-ethyl acetate-isopropanol-water(8:3:1.5:0.5).Separation were performed on a C_(18) (4.6 mm× 250 mm,5 μm) column of HPLC ;The mobile phase consisted of methanol-0.01 mol·L~(-1) potassium di- hydrogen phosphate(10:90)at the flow rate of 1.0 mL·min~(-1);the detection wavelength was 254 nm and column temperature was 26 ℃.Results:The calibration curve was in the range of 2.08 ng-2.08μg(r=0.9999) ;The av- erage recoveries of adenosine was 100.3% (n=9).Conclusion:The TLC and the HPLC methods are simple and reliable for the controlling of the quality of Ganoderma capsules.
