Abstract

Objective To study the expression of brain derived neurotrophic factor(BDNF)and its receptor TrkB in the brain tissue resected from patients with intractable mesial temporal lobe epilepsy(MTLE)and discuss their role in the pathogenesis.Methods Immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR)were used to test the expression of BDNF and TrkB in the surgically removed hippocampi and temporal lobe cortex tissues from 11 patients with intractable MTLE,and the results were compared with those of controls.Results Expression of BDNF protein and TrkB protein were significantly increased in temporal lobe cortex and hippocampal tissue resected from patients with intractable MTLE compared with that of controls(P0.01).Expression of BDNF mRNA and TrkB mRNA were significantly increased in temporal lobe cortex(P0.01)and hippocampal tissue(P0.05)resected from patients with intractable MTLE compared with that of controls.Conclusion The results indicate that the expression of BDNF and TrkB increases in the temporal lobe and hippocampus of patients with intractable mesial temporal lobe epilepsy,and it may play an important role in the generation and development of epilepsy.
