Abstract
Objective To analyze the relationship between malignancy of glioma and mRNA expression level of gene MT1-MMP and provide information for clinical diagnosis, therapy and prognosis of the evaluation. Methods The mRNA expression level of gene MT1-MMP in 34 gliomas and 7 normal brain tissues were detected with quantitative real-time RT-PCR and semi-quantitative RT-PCR. Results With Real-time RT-PCR method, the △△Ct value of normal brain tissue and grade II(low-grade)、III、IV high-grade glioma were 0.834±0.517, 4.063±2.309, 6.376±4.023,7.653±4.196,7.072±4.072 respectively. With semi-quantitative RT-PCR method, the relative value of normal brain tissue and grade II(low-grade)、III、IV 、high-grade glioma were 0.799±0.156, 1.142±0.158, 1.538±0.482,1.652±0.376, 1.600±0.421 respectively. There was the statistical significant difference between normal brain tissue and grade II、III、IV glioma, grade II and grade IV glioma, normal brain tissue and low-grade、high-grade glioma, low-grade and high-grade glioma. Conclusions The mRNA expression of gene MT1-MMP is up-regulated in glioma, and the mRNA expression of gene MT1-MMP increases with the malignancy of glioma.
